Abstract

PENGARUH PENGENCER DAN LAMA SIMPAN PADA 5 °C TERHADAP
KUALITAS SPERMA SAPI BRAHMAN

Penelitian ini dilakukan untuk mengetahui apakah terdapat pengaruh
pengencer dan lama simpan pada 5 °C terhadap kualitas sperma sapi Brahman.
Penelitian ini dirancang dengan Rancangan Acak Lengkap (RAL) faktorial 5 x 4.
Semen hasil koleksi diencerkan menggunakan berbagai level pengencer sitrat kuning
telur dan sitrat madu antara lain S1(SKT 100% + SM 0%), S2(SKT 75% + SM
25%), S3(SKT 50% + SM 50%), S4(SKT 25% + SM 75%), S5(SKT 0% + SM
100%), disimpan pada 5 OC dan selanjutnya diamati pada P1 = 0 jam, P2 = 2 jam, P3
= 4 jam dan P4 = 6 jam. Data yang diperoleh dalam penelitian ini kemudian dianalisis
menggunakan analisis variansi, jika terdapat perbedaan nyata dilanjutkan dengan uji
Duncan’s Multiple Range Test. Rerata motilitas S1: 28,83%; S2: 36,00%; S3:
38,67%; S4: 35,50%; S5: 21,08%, P1: 41,00%; P2: 38,40%; P3: 26,80%; P4:
21,86%. Rerata viabilitas S1: 52,67%; S2: 55,75%; S3: 59,25%; S4: 39,83%; S5:
34,41%, P1: 56,93%; P2: 51,73%; P3: 44,46%; P4: 40,40%. Rerata abnormalitas
spermatozoa S1: 13,67%; S2: 12,75%; S3: 11,58%; S4: 14,91%; S5: 16,08%, P1:
11,00%; P2: 12,67%; P3: 14,40%; P4: 17,13%. Berdasarkan dari hasil penelitian
dapat disimpulkan bahwa pengencer semen terbaik diperoleh pada perlakuan
pengencer S3 (sitrat kuning telur 50% + sitrat madu 50%) dan lama waktu
penyimpanan terbaik pada lama simpan 2 jam (P2).

ABSTRACT*
This study was conducted to know the influence of diluent and storage
duration at 5 °C on brahman bull sperm quality . This research was designed with
Completely Randomized Design (CRD) 5 x 4. Semen collection result was diluted
using various levels of yolk citrate dilator and citrate honey among others S1 (SKT
100% + SM 0%), S2 (SKT 75% + SM 25 %), S3 (SKT 50% + SM 50%), S4 (SKT
25% + SM 75%), S5 (SKT 0% + SM 100%), stored at 5 oC and subsequently
observed in P1 = 0 hours, P2 = 2 hours, P3 = 4 hours and P4 = 6 hours. The data
obtained in this study were analyzed using analysis of variance, if there was a
significant difference in the test continued by Duncan’s Multiple Range Test. Mean
of motility S1: 28,83%; S2: 36.00%; S3: 38.67%; S4: 35,50%; S5: 21.08%, P1:
41.00%; P2: 38,40%; P3: 26,80%; P4: 21,86%. Mean viability S1: 52,67%; S2:
55.75%; S3: 59.25%; S4: 39.83%; S5: 34,41%, P1: 56,93%; P2: 51,73%; P3:
44,46%; P4: 40,40%. Mean of spermatozoa abnormality S1: 13,67%; S2:
12.75%; S3: 11.58%; S4: 14.91%; S5: 16.08%, P1: 11.00%; P2: 12,67%; P3:
14,40%; P4: 17,13% . Based on the result of the research, it can be concluded that the
best cement diluent is obtained on the S3 dilution treatment (50% yellow citrate
citrate + 50% citrate citrate) and the best storage time in 2 hours saving time (P2).
